The active tetrameric glucose dehydrogenase from Bacillus megaterium is rapidly inactivated upon reaction with tetranitromethane. The inactivation is correlated with the nitration of a single tyrosine residue/subunit. The nitration does not influence the dissociation-reassociation process of the enzyme. The inactivation is prevented by the presence of NAD, AMP, ATP. The sequence around the nitrated tyrosine residue was determined and the residue was identified as Tyr-254 in the covalent structure of the enzyme. After dissociation of the enzyme into its monomers two tyrosine residues become susceptible to nitration. The nitrated subunits are unable to reassociate to the tetramer. Isolation and sequence analysis of the peptides containing nitrotyrosine indicated that two different tyrosine residues are predominantly modified. One residue is Tyr-254 which is essential for the catalytic activity and the other one is Tyr-160 which seems to be located in the subunit binding area. 相似文献

Summary We have cloned lamB, the gene for receptor (an outer membrane protein), on a small plasmid which also carries the gene for -lactamase (a periplasmic protein). We have identified a promoter in the region of malK, the gene immediately preceding lamB, which is active in minicells but relatively inactive in vitro. Using a minicell system, we have found that both receptor and -lactamase are made as full length precursors which are subsequently processed. We also show that the receptor precursor can be exported to the outer membrane before it is processed. Mature -lactamase is found only in the periplasm, suggesting that processing may be a requirement for export to the periplasm. 相似文献

The prenatal effects of mycotoxins were investigated in GBA mice given by stomach tube a single dose of either aflatoxin B1 (4 mg/kg), ochratoxin A (8 mg/kg) or zearalenone (20 mg/kg) on pregnancy day 8 or 9. Aflatoxin caused foetal anomalies (exencephaly, open eyes, and protrusion of intestines) after exposure on gestation day 8 but not on day 9. The effects (increased prenatal mortality, reduced foetal growth, and a wide variety of malformations) caused by ochratoxin were much more severe and occurred after treatment on either of the 2 days of gestation. Among the spectrum of malformations, predominantly involving the craniofacial complex and the axial skeleton, the most striking was the total aplasia/dysplasia of the upper facial structures. These defects were always accompanied by exencephaly and anophthalmia. Zearalenone caused no effects. It is concluded that of the 3 mycotoxins screened with the technique used, ochratoxin is the most potent teratogen in mice. 相似文献

Summary 22 independent man-hamster (HGPRT–) hybrids using male human cells with balanced reciprocal translocation t(X;2)(p22;q32) were analysed for human genes localized on chromosome 2 (IDHS, MDHS), on chromosome X (PGK, GAL, G6PD) and for the different chromosomes in relation with the balanced reciprocal translocation (chr.2, chr.2q–, chr.Xp+).The following results were obtained:The chromosomes 2 and 2q– are absent in the 22 hybrids.In 9 hybrids, the absence of MDHS in spite of the presence of the chromosome Xp+ indicates that the gene for MDHS is not localized on this chromosome (or that the gene for MDHS is not on the segment 2q32-2qter translocated on X).In 14 hybrids, the three markers of X (PGK, GAL, G6PD) and IDHS are expressed in the presence of the chromosome Xp+. This result indicates that the genes for these markers are on Xp+ or that the genes PGK, GAL, G6PD are on X without the Xp22-Xter segment, translocated on the chr.2, and that the gene for IDHS is on the 2q32-2qter segment translocated on X.In 8 hybrids, in the absence of the intact chromosome Xp+, the higher percentage of the presence of G6PD (7 hybrids) and the lower percentage of the presence of IDHS (3 hybrids) are explained by the fact that these hybrids selected in HAT medium had to retain a segment of Xp+ bearing the human gene HGPRT. G6PD appeared very close to HGPRT and IDHS very distant from HGPRT.The study of the different correlations between the presence and the absence of these four markers on Xp+ in the different hybrids indicates the following order on the chromosome Xp+ from p to q: IDHs — PGK — GAL — G6PD.

Characterization and classification of virus particles associated with hepatitis A. I. Size, density, and sedimentation. 下载免费PDF全文
Virus-like particles were purified from stools of patients in an epidemic of hepatitis A in Germany. When reference MS-1 chimpanzee pre-inoculation and convalescent sera were used, the close serological relationship of the purified particles to well-known isolates of hepatitis A could be established. On the other hand, the physicochemical characteristics of the particles were determined in parallel to the characteristics of a marker parvovirus (LuIII) and a marker picornavirus (poliovirus type 2). It could be shown that the majority of the hepatitis A-associated particles band at 1.34 g/ml in CsCl and, like poliovirus, sediment at about 160S. In addition, a distinct hepatitis A antigen was observed, which banded at 1.305 g/ml and sedimented between 50 and 90S. A further component accumulated in the density range of between 1.38 and 1.44 g/ml. However, it seemed to be rather labile. Upon reisolation from CsCl and sedimentation in sucrose, it resolved into a 160S, a 90 to 100S, and a 50S form. The size of the 160S particles (27 to 29 nm) could be readily distinguished from that of the parvovirus (22 to 24 nm). It is concluded, therefore, that hepatitis A-associated virus particles are more likely to be classified with the picornaviruses than with the parvoviruses. 相似文献

A low IgG2a response in B10 mice during the primary response to sheep red blood cells (SRBC) is described. Analysis of the response in B10 × BALB/c hybrid progenies and in congenic strains indicates that this low response is a dominant phenotype placed under the control of a single Mendelian gene or a group of closely linked genes. Based on SEM examinations of some 1600 species of monocotyledons out of 61 families the characteristics of their epicuticular waxes are described in this preliminary paper and illustrated by 20 SEM micrographs. Four basic types of waxes are recognized. Two of them occur throughout the monocotyledonous and dicotyledonous orders of angiosperms; two others are of high systematic significance and restricted to particular orders of the monocotyledons: The “Convallaria-Type” is characterized by fine wax platelets with parallel orientation, forming a pattern around the stomata reminiscent of electromagnetic field lines. This type is restricted to the three liliiflorous ordersAsparagales, Liliales, andBurmanniales. The binding of 4-methylumbelliferyl-α-D-galactopyranoside, -β-D-galactopyranoside and -D-Galβ(1→3)DGalNac to peanut agglutinin was studied by fluorescence. Peanut agglutinin quenched the fluorescence intensity of 4-methylumbelliferyl-α-D-galactopyranoside but enhanced that of the two 4-methylumbelliferyl-β-galactosides. For α-D-galactopyranoside, the association constants measured at 4 and 25°C were 3.4 × 103 and 1.7 × 103 M?1 respectively, and for D-Galβ(1→3)DGalNac, 1.5 × 105 and 3.3 × 104 M?1. The binding enthalpies estimated from these values are consistent with the existence of extended sugar binding sites in the peanut agglutinin molecule. 相似文献
